Uncouplers of Oxidative Phosphorylation
نویسنده
چکیده
was proportional to the acidity of the imidazole -NH group. Tetrachloro-2trifluoromethylbenzimidazole was the most active (50% uncoupling of oxidative phosphorylation at 7-9 x 10-8M, pK5.04). Nitro-substituted analogues were less active than predicted from pK considerations or from partition-coefficient measurements. 3. Introduction of an -NH2 or -CO2H substituent caused a loss of uncoupling activity, as did alkylation at position 1 of the imidazole ring. 4. Benzimidazoles active as uncouplers stimulated mitochondrial adenosine triphosphatase but not all stimulated the oxidation of succinate in the absence ofa phosphate acceptor. 5. 4,5-Dichloro-2-trifluoromethylbenzimidazole inhibited the succinate-oxidase system at about the same concentration required for uncoupling (0.52Mm for 50% inhibition of both activities) and the site of this inhibition appears to lie between succinate dehydrogenase and cytochrome b.

Uncouplers of oxidative phosphorylation.
Uncouplers of oxidative phosphorylation in mitochondria inhibit the coupling between the electron transport and phosphorylation reactions and thus inhibit ATP synthesis without affecting the respiratory chain and ATP synthase (H(+)-ATPase). Miscellaneous compounds are known to be uncouplers, but weakly acidic uncouplers are representative because they show very potent activities. The most poten...
متن کاملEnergy coupling in the transport of beta-galactosides by Escherichia coli: effect of proton conductors.
Escherichia coli accumulates thiomethyl-beta-d-galactoside against a concentration gradient under anaerobic conditions. The accumulation was abolished by carbonylcyanide m-chlorophenylhydrazone, tetrachlorosalicylanilide, 2,4 dinitrophenol, and other uncouplers of oxidative phosphorylation even though oxidative phosphorylation would not be expected to occur anaerobically. In the presence of the...
متن کاملStudies on the Influence of Various Metabolic Uncouplers on the Biodegradation Rate of Toluene in a Biofilm Bio-Filter Reactor
Biological inhibition of air pollution has vast advantages over physicochemical methods. One of the biggest challenges faced by researchers with traditional bio-filter in controlling Volatile Organic Compounds (VOCs) such as Benzene, Toluene, Ethylbenzene, and Xylene (BTEX) is, low degradation rate (elimination capacity) and accumulation of very high biomass. The use of metabolic uncouplers...
متن کاملRecognition sites for chemotactic repellents of Bacillus subtilis.
Repellents of Bacillus subtilis include many membrane-active compounds, such as uncouplers of oxidative phosphorylation, local anesthetics, chlorpromazine (a central nervous system depressant), and tetraphenylboron (a lipophilic anion). Normally, bacteria swim smoothly, and occasionally tumble, but addition of repellent causes all bacteria to tumble, then later resume original frequency of swim...
متن کامل6-ketocholestanol abolishes the effect of the most potent uncouplers of oxidative phosphorylation in mitochondria.
The effect of a keto-derivative of cholesterol, namely, 6-ketocholestanol (5 alpha-cholestan-3 beta-ol-6-one; kCh) on the uncoupling of oxidation and phosphorylation by various uncouplers was studied in rat heart mitochondria. kCh was found to completely abolish the uncoupling effect (the increase in the respiration rate under the respiratory control conditions and the decrease in the membrane ...
متن کاملIsolation and reactions of a phosphorylated form of phosphoryl transferase from beef heart mitochondria.
Phosphoryl transferase, a mitochondrial protein which increases the phosphorylative capacity of poorly phosphorylating submitochondrial particles and catalyzes an ATP-ADP exchange reaction, is phosphorylated during oxidation either of succinate or pyruvate-malate. Inhibitors of oxidative phosphorylation and electron transfer, as well as uncouplers of oxidative phosphorylation, inhibit the phosp...
